LowCross-Buffer®使用常见问题及解答

1. LowCross-Buffer®具体是什么?

LowCross Buffer®是一种样品/抗体缓冲液，用于免疫分析（如ELISA、Western blotting、蛋白质阵列和免疫PCR）。它有助于消除干扰。干扰如非特异性结合、交叉反应和基质效应。抗体和抗原之间的高亲和力结合不会受到负面影响，而抗体的不必要结合会受到抑制。

2. LowCross-Buffer®为什么能减少基质效应?

大多数所谓的基质效应都是基于基质成分（如人血清样本中的血清蛋白）与待测物或抗体之间不需要的低或中亲和力结合。

待测物可能会被样本中的蛋白质或其他成分所掩盖，从而导致抗体不能与待测物结合。此外，基质成分也可能会掩盖抗体。LowCross Buffer®减少了抗体的掩蔽。

3. LowCross-Buffer®该怎样使用?

LowCross Buffer®是用于标本（如血清或血浆）和/或检测抗体的稀释缓冲液。这取决于实验干扰。如果干扰如非特异性结合或交叉反应来自检测抗体，我们建议在LowCross Buffer®中稀释检测抗体。

4. LowCross-Buffer®需要稀释使用吗?

不用。LowCross-Buffer®为即用型，

样本或检测抗体可直接用这个缓冲液稀释。只有在竞争性分析或者某些应用如免疫组织化学中，可以用水货PBS稀释 LowCross-Buffer®后使用。在这种情况下，用户需通过测试找到理想的混合比。

5. LowCross-Buffer®可以与单克隆或多克隆抗体一起使用吗?

可以。但如果你使用多克隆抗体，需注意多克隆抗体组成中含有许多不同亲和力的抗体，LowCross-Buffer®仅允许高亲和力结合，而低亲和力和重亲和力结合则会减少。如果使用LowCross-Buffer®，可能需要适度增加多克隆检测抗体的浓度。抗体浓度的增加也会增加高亲和力抗体的浓度。而LowCross-Buffer®可以抑制低亲和力和中亲和力抗体的结合。

是否必须增加抗体的浓度取决于抗体的质量。

6. LowCross-Buffer®缓冲液的组分是否会影响酶法检测的显色反应?

LowCross-Buffer®不会对碱性磷酸酶或过氧化物酶的酶活性产生负面影响。有时用户使用LowCross-Buffer®孵育标记的抗体可以增加酶活性。

7. 采用过氧化物酶标记二抗进行检测的夹心法ELISA，实验结果出现背景的情况，这是什么原因导致的呢？使用LowCross-Buffer®是否会有帮助?

重要的是要知道二抗是否经其他哺乳动物的抗体预吸附处理。抗兔IgG的二抗也可与小鼠IgG或山羊IgG发生高亲和力的交叉反应，除非针对这些物种的抗体进行纯化。原因是哺乳动物抗体序列同源性高。二抗通常不纯化（预吸附），因此会发生高亲和力的交叉反应。在这种情况下，使用LowCross Buffer®没有帮助。二抗必须更换成不与包被抗体发生交叉反应的预吸附的抗体。

8. LowCross Buffer®是否可以与荧光染料结合使用，譬如在蛋白分析中？

可以。蛋白分析中会带来积极的实验结果。

在某些使用青色染料（例如Cy5、Cy3、Oyster-dyes) 应用中，结合LowCross Buffer®使用可以观察到荧光信号增强。

9. LowCross-Buffer®能否多次冻融?

LowCross-Buffer®可经数次冻融，但重要的是在使用前需将该缓冲液充分混合。

10. LowCross-Buffer®能否替代如Western blotting膜、ELISA板的表面封闭剂?

不可以! LowCross-Buffer®不是表面封闭剂，你可以用她稀释样本或抗体。对于封闭，我们建议使用The Blocking Solution。这款封闭剂的封闭效果比市面上大多数封闭剂的要更优。

聚苯乙烯或玻璃表面包被抗体和抗原的稳定剂

固定抗体/抗原并封闭后，Liquid Plate Sealer® 用均匀的稳定层密封板，显示出良好的溶解性，并且不影响之后的测定。

它还可用于在蛋白质阵列的聚苯乙烯或玻璃表面涂覆后稳定抗体和抗原。用 Liquid Plate Sealer® 处理的印版在干燥后可以长期储存。

即使在室温下，也可以在干燥条件下长期储存涂层板。

推荐的最佳储存温度为 2 – 8°C。但大多数稳定的板也可以在环境温度下干燥储存数月而不会失去活性。
