Her-2 CTC在液相中的免疫荧光染色（用于小鼠样品中人类CTC的基因组分析）

描述

通过抗-Her2免疫荧光染色鉴定从Her-2阳性实体瘤如乳腺癌或胃癌中脱落的富集的Her-2阳性CTC。抗Her2与Her-2 CTC结合的证明提供了关于评估抗Her2治疗功效的更多见解。可以根据需要同时进行针对额外的上皮细胞，内皮细胞和干细胞等标记的抗体。

从液体中富集的人类CTC的免疫荧光染色容易，快速。鉴定的细胞具有较亮的荧光强度，可用于小鼠样品中人类CTC的基因组分析。

溶液配制

1.储备溶液配制

所有未使用的储备溶液应分为一次性使用的等分试样，并在制备后储存在-20°C下。避免重复冻融循环。
1. CTC储备溶液（10倍）：
将1 mL蒸馏水加到CTC（组分A）小瓶中，制成10X储备液。注意：原液在-20°C稳定2周。避光。

操作步骤

1.准备细菌样品，浓度为10⁶细胞/ ml。在适当的培养基中使细菌生长到对数后期。通过以10,000 x g离心10分钟除去培养基，然后将沉淀重悬于适量的PBS中。注意：在波长= 600 nm（OD600）处测量细菌培养物的光密度，以确定细胞数。对于大肠杆菌培养，OD600 = 1.0等于8 x 10⁸细胞/ ml。

2.根据需要用测试化合物处理细胞。通过以10,000 x g离心10分钟来去除处理液，然后将沉淀物重新悬浮在适量的测定缓冲液（组分B）中，以使处理后样品中的细菌浓度与活菌浓度相同。注意：开始治疗之前，请确定细菌培养物的浓度。注意：死细菌可以作为阴性对照。建议用70％乙醇杀死细菌30分钟，然后煮沸60分钟。

3.将10 µL 10X CTC储备溶液加到90 µL细菌样品中。

4.充分混合，然后在黑暗中于37°C孵育30分钟。

5.在使用流式细胞仪分析细胞之前，添加400 µL分析缓冲液（组分B）以增加体积。

6.用流式细胞仪通过PE-Taxes Red通道（Ex / Em = 488/615 nm）监测细菌的荧光。注意：要排除杂物，建议将流式细胞仪的阈值设置为以下值：FSC> 10,000，SSC> 5,000。注意：CTC的效率高度依赖菌株，因此会相应优化染色条件。
