内皮素受体B在小鼠JG细胞中的表达。使用抗内皮素受体B抗体（＃AER-002）（红色）对小鼠离体的肾小球（JG）细胞进行免疫细胞化学染色。ET-B染色与肾素（肾细胞标记物）共定位（绿色）。改编自Ortiz-Capisano，MC （2014） Physiol。代表 2， e12240。经美国生理学会和生理学会的许可。

抗内皮素受体B抗体（＃AER-002）是针对大鼠ET-B表位的高度特异性抗体。该抗体可用于蛋白质印迹，免疫细胞化学，免疫组织化学和间接流式细胞仪应用。它被设计为从大鼠，小鼠和人类样品中识别ET-B。

免疫组织化学

EDNRB在大鼠肺中的表达

使用抗内皮素受体B 抗体（＃AER-002）（1：100）对大鼠肺部石蜡包埋的切片进行免疫组织化学染色。EDNRB在呼吸道上皮（黑色箭头）和呼吸道平滑肌（红色箭头）中都有表达。请注意，血管平滑肌未染色（蓝色箭头）。苏木精用作复染剂。
EDNRB在大鼠丘脑中的表达

使用抗内皮素受体B 抗体（＃AER-002）对大鼠背侧脑室周围丘脑进行免疫组织化学染色。甲酚紫用作复染。

形成冻干粉末。重建的抗体包含pH 7.4的磷酸盐缓冲盐水（PBS），1％BSA，0.05％NaN 3。

同种兔IgG。

每个批次的标准质量控制Western blot分析。

肽确认通过氨基酸分析和质谱确认。

重构前的储存抗体在室温下以冻干粉的形式运送。到达后，应将其保存在-20°C下。

根据样品量，重新配制25 µl，50 µl或0.2 ml的双蒸馏水（DDW）。

重构后的储存重构后的溶液可以在4°C下保存1周。对于较长的时间，应将小等分试样在-20°C下储存。避免多次冻结和解冻。使用前将所有抗体制剂离心（10000 xg 5分钟）。

重建前阴性对照抗原的储存冻干粉可在室温下完整保存2周。对于更长的时间，应将其保存在-20°C下。

阴性对照抗原重建100 µl双蒸馏水（DDW）。

-20°C复溶后阴性对照抗原储存。

前吸附对照每1 µg抗体1 µg肽。

科学背景

内皮素（ET-1、2和3）被认为是非常强大的血管收缩物质。在人类中，内皮素通过两个特定的G蛋白偶联受体ET A R 和ET B R 介导其作用。ET A R和ET B R在心脏和人心肌中的含量相似。1,2

内皮素受体的配体特异性不同。尽管ET A R对内皮素同工型具有不同的亲和力（ET-1> ET-2> ET-3），但ET B R没有显示出选择性亲和力。2,3

随后的研究表明内皮素在具有多种生物活性的血管以及非血管，细胞和组织中均存在。

目前，越来越多的证据表明，ET-1可能调节有丝分裂，凋亡，血管生成，肿瘤侵袭和转移的发生。3

直到近，人们还认为内皮素的所有细胞活性都是通过它们与细胞表面受体的相互作用来介导的。但是，近的一项研究表明，心脏核还具有ET A R和ET B R亚型，它们具有功能并与核膜内的信号传导机制偶联。2

在几种前列腺，膀胱和结肠癌细胞系中观察到ET B R的甲基化与转录下调和ET B受体表达降低有关。3

原生Psalmotoxin1（PcTx1）是一种40个氨基酸的毒素，zui初分离自南美洲狼蛛弓形猴的毒液。¹

PcTX1的特征在于异常的四联体Lys ²⁵ -Arg ²⁶ -Arg ²⁷ -Arg ²⁸，其可能在毒素分子的表面形成强阳性“斑块”，构成通道结合位点识别的强候选区域。PcTX1的分子支架可能类似于先前描述的锥形蜗牛和蜘蛛毒素^5,6，并且包括由三个二硫键网格化的三链反平行β-折叠结构。¹

PcTx1（IC ₅₀ = 0.9nM ），并且特异性阻断H ⁺阳离子通道的特定亚类，酸感应离子通道1a（ASIC1a），并且能够区分两个接头变体亚型，并且不阻断ASIC1b通道（仅在其N端序列上不同）。此外，PcTX1一旦该子单元与另一个家族成员（ASIC2a或ASIC3）相关联，就失去了阻止ASIC1a的能力。¹

ASICs在脑和脊髓神经元丰富，涉及疼痛感觉，缺血性脑卒中机械补偿，学习和记忆。^2,3 在急性和神经性疼痛模型中，ASIC1a与PcTx1的特异性阻断导致在阿片受体上游工作的镇痛作用。⁴

使用碘化形式的毒素，PcTX1位点的结合鉴定了ASIC1a细胞外环的富含半胱氨酸的结构域I和II（CRDI和CRDII）。通道的区域1和4之间的二硫桥支持在其中PcTx1作用的CRDI4-CRDII结构域与质子⁷的结合位点的后M1区之间的紧密关系。

Alomone Labs Psalmotoxin-1抑制在非洲爪蟾卵母细胞中表达的ASIC1a的阳离子电流。
