可用的TLC 背板TLC板中

1、玻璃背板TLC板TLC中常使用的支撑是玻璃板,因为它具有耐化学性和易于处理板材。玻璃板的缺点是其易碎性,尺寸和重量(玻璃板厚约1.3mm)并且需要额外的保护性包装材料。

2、塑料背板TLC板:聚酯板(约0.2毫米厚)具有以下几个优点:它们可以抵抗所有常见溶剂,需要更少的填料,并且需要更少的储存空间。的塑料背衬的TLC板可以用剪刀轻易切割成任何所需的尺寸和斑点可以被切割和洗脱。炭化技术可用于二氧化硅涂覆的塑料TLC板。炭化必须在比玻璃更低的温度下进行。塑料TLC板的高温度为160°C。

3、铝背板TLC板:由于铝支撑较厚(约0.15 mm厚),铝背板TLC板玻璃或塑料支撑的TLC板更稳定,更容易切割。即使是20 x 20厘米的铝背板TLC板,其抗扭强度几乎与玻璃板一样好。铝背衬TLC层具有出色的粘附性,可以将板材切割成任何所需尺寸,而不会干扰吸附剂层。使用与我们的TLC玻璃板相同的经过时间测试的粘合剂系统涂覆普通铝箔支撑体。对于含有高浓度水的洗脱液,我们建议使用预涂铝TLC板。

塑料背衬的TLC板片一样,玻璃和铝TLC板可以用纯水性可视化试剂喷涂或浸渍,而不会对层造成任何明显损坏。使用铝背板TLC板时,使用含有高浓度无机酸或浓氨水(与铝反应)的洗脱液时可能会出现问题。作为背衬不稳定问题的解决方案,我们建议使用我们的玻璃或聚酯TLC板铝背板TLC板具有与塑料背衬TLC板相同的优点,具有更高的耐温性(例如用于炭化技术)。

离子交换PhyTip色谱柱

PhyTip ®离子交换柱提供了在一次实验中筛选不同洗涤和洗脱缓冲液以及许多树脂的*能力。一旦优化了离子交换条件,它们就可以直接转移到更大的生产规模的色谱柱。使用来回流动在移液管柱中进行色谱分离以驱动结合并释放平衡步骤。使用MEA 2个人净化系统可以一次处理12个样品。使用机器人液体处理系统可以将其增加到96个样品。PhyTip色谱柱有四种标准离子交换树脂(强阴离子,弱阴离子,强阳离子,弱阳离子)。对于树脂选择,离子交换树脂筛选工具包是一个包含24根每种树脂的96个的盒子。

离子交换的一个优点是用于捕获洗涤和洗脱的缓冲条件通常与蛋白质相容,从而可以在终产物中保留高活性。此外,离子交换色谱仍可用于终纯化和抛光。使用离子交换作为亲和色谱的正交化学可以产生非常纯的蛋白质。

有两种基本类型的离子交换剂:阳离子交换剂和阴离子交换剂。阳离子交换分离在pH为4-7之间进行,阴离子交换分离通常在pH为5-8之间进行。

在阴离子交换中有两种类型:强碱阴离子交换和弱碱阴离子交换。阳离子交换也是如此:强酸阳离子交换和弱酸阳离子交换。

选择有效的离子交换剂和离子交换条件可能难以确定离子交换纯化。基于移液管柱的PhyNexus PhyTip色谱柱技术可以轻松地并行处理样品,从而可以在很短的时间内有效地筛选数十到数百个离子交换。

离子交换柱采用我们专有的  双流色谱,可  提供完整的抗体结合和高终浓度。PhyTip色谱柱兼容大多数8,12和96通道自动化液体处理机器人,包括  Agilent Technologies,  Beckman Coulter,  Dynamic Devices,  Perkin Elmer,  Hamilton和  Tecan。

PhyNexus的PhyTip产品系列包括离子交换。在传统的离子交换色谱中,样品,洗涤缓冲液和梯度洗脱缓冲液在一个方向上通过填充柱。PhyTip色谱柱能够通过驱动结合和释放步骤达到平衡,在一个小的移液管柱中模拟这种色谱分离。这不是通过使用高背压,大柱,而是通过PhyNexus获得专项技术的来回流动来实现的。
