组织型纤溶酶原激活剂，又称组织型纤溶酶原激活物（tissue—type plasminogenactivator，t-PA）或纤溶酶原激活因子（tissue plasminogen activator），是体内纤溶系统的生理性激动剂，在人体纤溶和凝血的平衡调节中发挥着关键性的作用。属于糖蛋白,丝氨酸蛋白酶类。是一种新型的血栓溶解剂。由于血栓性疾病的逐年上升，对溶栓药物的需求量逐年增加，使对溶栓药的研究成为热点，而目前又以对tPA及其突变体、嵌合体的研究zui多。

尿激酶型纤溶酶原激活剂选购指南

尿激酶型纤溶酶原激活物(Urokinase type Plasminogen Activator (uPA) )，uPA是一种丝氨酸蛋白水解酶。小鼠uPA基因位于第10条染色体长臂上，其长度为6.5KD，由11个外显子构成，可转录成24KB的成熟的mRNA，初始翻译产物为前尿激酶原（含431个氨基酸残基），除去20个氨基酸残基的信号肽后即为细胞分泌的尿激酶原（pro-uPA），在158LYs位处劈裂，则成为由二硫键连结的双链活性型结构，即20×103的A链和34×103的B链。C端的A链包括一个Kringle区和表皮生长因子（EGF）功能区，表皮生长因子功能区能介导与特异的受体结合，去掉此功能区，其仍具有激活纤溶酶原作用，,但却无与受体结合的能力。C端的B链有丝氨酸蛋白酶功能区，为uPA的活性中心。uPA的合成与分泌受诸多因素影响，如原癌基因V-Src、Ras、细胞因子TNF、INF、IL-1、TGF-β、IGF-1、FGF、基质金属蛋白酶等。一般说来，uPA主要在生理、病理条件下与细胞分化、迁移、组织重建、细胞周围基质降解有关。

Purified from homogeneous glu-plasminogen by activation with Plasmin. This activation results in the release of a 76 amino acid residue peptide （Glu-Lys76）. This Lys77-Plasminogen can be readily converted to Lys77-Plasmin by any of the common plasminogen activators. The protein purity is determined by SDS-PAGE.

Purified from fresh frozen human plasma. The protein purity is determined by SDS-PAGE and shows no reduction upon incubation with 2-mercaptoethanol. No Plasmin activity is detected using the chromogenic substrate S-2251. Plasminogen is activated to the serine protease plasmin via urokinase, streptokinase or tissue plasminogen activator.
